“ Our results from Drosophila with a deeper population sample lend credence to the hypothesis that, in humans too, a force of strong constraint is responsible for the lack of polymorphism at 4D sites rather than a mutational force or other confounding factors. For many species, there has been no conclusion that the constraint on their respective synonymous sites is strong, but many of the signals are consistent with what we find in Drosophila with the fraction of sites under constraint, the amount of missing polymorphism, and the lack of relationship to codon bias. ”
