Idioplasm

“ Every cell contains idioplasm, since every cell-nucleus contains chromatin, but I call the idioplasm of the germ-cells germ-plasm, or the primary-constituent-substance of the whole organism, and the complexes of primary constituents necessary to the production of a complete individual—whose presence we have just shown to be theoretically necessary—I call ids. In many cases these 'ids' might be synonymous with chromosomes, at least in all the cases in which the chromosomes are simple, that is, are not composed of several similarly formed structures. ”

“ It appears to me to be a necessary consequence of Nägeli’s theory that the causes of transformation lie solely in this molecular structure of the idioplasm. If nothing more than a certain amount of growth, and consequently a certain period of time during which the organism reproduces itself with a certain intensity, is required to produce a change in the idioplasm, then we must conclude that the alteration in the latter must take place when this certain amount of growth has been reached, or after this certain period has elapsed. ”

“ Each ontogeny (individual) begins in a minute germ cell, in which a small quantity of idioplasm is contained. In the cell divisions, by which the organism grows, the idioplasm divides into as many parts as there are single cells, while it continually increases in quantity in a corresponding degree. The ontogenetic increase of the idioplasm takes place by length growth of the strands—that is, by intercalation of micellæ in each row of cells of the strands, which thereby grow in length without changing the configuration of the cross section. ”

“ The time required by the idioplasm to undergo such changes as would constitute transformation into a new species ought to be always the same at equal heights in the scale of organization, that is, with equal complexity in the molecular structure of the idioplasm. ”

“ If heredity and variation are defined according to the true nature of organisms, they are only apparent opposites. Since idioplasm alone is transmitted from one ontogeny to the next following, the phylogenetic development consists solely in the continual progress of the idioplasm and the whole genealogical tree from the primordial drop of plasma up to the organism of the present day (plant or animal) is, strictly speaking, nothing else than an individual consisting of idioplasm, which at each ontogeny forms a new individual body, corresponding to its advance. ”

“ External stimuli impign upon the organism usually at a definite point, but they not only effect a local transformation of the idioplasm but also reproduce themselves in a dynamic manner in the entire idioplasm, which is in unbroken connection throughout the whole individual. The idioplasm is thus changed everywhere in the same manner, so that the germ cells that are given off at any point feel and inherit the effects of those local stimuli. ”

“ The idioplasm is the physiological equivalent of this specific individual quality of the sensations and perceptions, and there are reasons analogous with those in favour of the supposition of an idioplasm for the supposition of an individual character amongst animals. The sportsman who has to do with dogs, the trainer with horses, and the keeper with animals will readily admit the existence of this individuality as a constant element. It is clear that we have to do here with something more than a mere rendezvous of perceptions. ”

“ Unfortunately, Weismann's hypothesis tells us nothing at all about these internal causes, that depend upon the physical nature of the idioplasm; that is to say, nothing at all about the causes which, working in a fashion so contradictory and astonishing, really produce the whole development.
In such a state of affairs it is better to turn to Nature herself; and to see whether or no the occurrence of differentiating division of the nucleus in the organic world is at all supported by the actual observations and investigations of those who study cells. ”

“ If we take a survey of the assumptions we have been compelled to make from the standpoint of the theory to explain the development of germ-cells, budding, and regeneration, it would seem as if it were contradictory to assume that, on the one hand, complete germ-plasm should be given to certain cell-series as inactive accessory idioplasm, and, on the other, that very numerous cells, at least in the lower Metazoa, should have received the idioplasm of budding, and still more numerous cells that of regeneration. ”

“ The idioplasm continually alters its configuration with its growth in successive ontogenies, but comparatively very slowly, so that it makes a minute advance from the germ of one generation to the germ of the next. The summation of these increments of advance through a whole line of evolution represents the race history of an organism, since the latter is connected only by its idioplasm in unbroken continuity with the micellar beginning of its race. ”

“ Accordingly, each strand of idioplasm contains all the determinants that the particular individual has inherited in the germ cell, and each cell of the organism is idioplasmatically qualified to become the germ cell of a new individual. Whether this qualification may be realized depends upon the nature of the soma-plasm. In the lower plants this power belongs to each individual cell; in the higher plants many cells have lost it; in the animal kingdom it is possessed in general only by cells normally set apart as asexual or sexual reproductive cells. ”

“ These variations are inheritable in the phylogenetic sense and contribute to the formation of varieties and species; they always appear as the results of more or less secondary reactions which make their appearance with stimuli exerted by external causes.
External stimuli exerted on the organism are reproduced in the idioplasm. Since the stimulus is discontinued with each change of the ontogeny and only the idioplasm persists, permanent variations are produced only in the idioplasm by those conditions that produce visible transformations in the mature organism. ”

“ But even if it is theoretically possible to imagine that idioplasm possesses such a structure that it changes in a certain manner, as the result of mere growth, we should not be justified in thus assuming the existence of a new and totally unknown principle until it had been proved that known forces are insufficient for the explanation of the observed phenomena. ”

“ But if we admit a real change in constitution, then the backward development of the idioplasm of the cells of the body into germ-cells appears to be very far from a matter of course, and he who assumes it must bring forward weighty reasons. ”

“ Although in many cases the cell-bodies of such early embryonic cells fail to exhibit any visible differences, the idioplasm of their nuclei must undoubtedly differ, or else they could not develope in different directions. ”

“ Races arise from gamogenic or pathological variations of the idioplasm. In the former case they presuppose crossing between related varieties or species, in the latter case an increased sensibility and weakening of the idioplasm. Very often both causes co-operate, since crossing follows more easily when the idioplasm is weakened by hurtful influences and since the irritability and weakening of the idioplasm increases if crossing has preceded. Race formation begins in single individuals. ”

“ In the formation of the germ cells in sexual reproduction, the idioplasms of both parents must come into contact with each other, whereupon there results either a material union and formation of a mixed idioplasm or perhaps rather a dynamic action ”

“ Since, according to Nägeli, new life with new idioplasms, may arise wherever and whenever the necessary conditions combine, the present organic world is not made up from branchings of a single original idioplasm, but each race or group may have its own specific idioplasm ”

“ Thus the organism must contain at least as many determinants in its idioplasm as there are different phenomena in its inheritable ontogeny; and if new phenomena appear in it, new clusters of micellæ must previously have been introduced into the idioplasm, or the orientation and arrangement of clusters already present must have been changed. The formation of such a determinant, whether it concerns the perfecting of the organism or its adaptation to environment, always proceeds very slowly, and as a rule has no effect before its completion. ”

“ The theory of an idioplasm, the presence of which gives the specific race characters to those tissues and cells which have lost the reproductive faculty, is by no means generally accepted. But at the least all must admit that the race characters are collected in the genital glands, and that if experiments with extracts from these are to provide more than a good tonic, the nearest possible relationship between the animals experimented upon must be observed. ”

“ On the other hand, the principle of organotherapy has been opposed on the ground that organic preparations procured from non-allied species could not possibly be expected to yield good results. It is more than likely that the medical exponents of organotherapy have lost many valuable discoveries in healing because of their neglect of the biological theory of idioplasm. ”

“ The species arises neither from the nutrition variety nor from the race; it is always a more advanced variety, and hence species formation is identical with variety formation. Cause for variation and consequently for variety formation is always shown, either when, environment remaining the same, the automatic variation of the idioplasm has advanced so far that the ontogeny is raised to a higher grade of organization and division of labor, or when external stimuli act for a sufficiently long time in a manner not in harmony with the previous adaptation. ”

“ Idio-plasm-B is the substance out of which future cells are to be formed and controlled, when in due course either of ontogeny or phylogeny this idio-plasm-B becomes converted into idio-plasm-A,—i.e., into each subsequently developing tissue or organism, as the case may be. I say ontogeny or phylogeny, and tissue or organism, because, where a germ-cell is concerned, idio-plasm-B is capable of reproducing entire organisms of its own and of subsequent generations ”

“ The nucleus stains very deeply with all dyes; the protoplasm possesses a strong attraction for most acid stains, and is unmistakeably characterised by the presence of a dense neutrophil granulation. The reaction of the protoplasm is alkaline, to a less degree however than in the lymphocytes. No free glycogen is contained in the polynuclear cells as a rule; nevertheless in certain diseases cells are always found which give a marked iodine reaction. In this manner the appearance of cells containing glycogen in diabetes was first proved. (Ehrlich, Gabritschewsky, Livierato.) ”

“ Perhaps the most remarkable class of stimulations, however, is that due to the presence of the entire protoplasmic body of one organism in the cell of another, each living its own life for the time being, but the protoplasm of the host cell showing clearly, by its abnormal behaviour, that the presence of the foreign protoplasm is affecting its physiology. A simple example is afforded by Zopfs' Pleotrachelus, the amoeboid protoplasmic body of which lives in the hypha of Pilobolus, causing it to swell up like an inflated bladder, in which the parasite then forms its sporangia. ”

“ Isn't this a marvellous picture? To see the micro-organism move, evolve and revolve in the midst of normal cells, uncoil and undulate in the fluids which they inhabit, to see them play hide and seek with the blood corpuscles and clumps of fibrin, turn, twist, and rotate as if in a cage, to see these deadly little trypanosomes moving back and forth in every direction displaying their delicate undulating membranes and shoving aside the blood cells that are in their way while by their side the leucocytes, or white corpuscles, lazily extend or retract their pseudopods of protoplasm. ”

“ It is not easy to understand why growth should have been less rapid here than elsewhere, as the position does not seem to correspond to any feature in the embryo. In some specimens the hypoblast cells at the extreme edge of the pellucid area are smaller than the cells immediately internal to them. At about the twenty-third hour these cells begin rapidly to lose the refractive spherules they contained in the earlier stages of incubation, and come [Pg 38] to consist of a nucleus surrounded simply by granular protoplasm. ”

“ Ectoderm cells, and possibly also some primitive endoderm cells, also take part in forming the test. Many of these cells in the test remain small and simple, as the fusiform and stellate test-cells; some become pigment-cells, while others enlarge and become vacuolated to form the large (up to 0.15 mm. in diameter) vesicular or "bladder" cells—this is especially the case in the outer layer of the test in Ascidia mentula (see Fig. 17, bl) where there are innumerable clear vesicles, each surrounded by a thin film of protoplasm and having the nucleus still visible at one point of the surface. ”

“ To their view, we cannot fancy protoplasm as so much matter in a pot, in an ointment-box, any portion of which scooped out in an ear-picker would be so much life-stuff, and, though a part, quite as good as the whole. This seems to be Mr. Huxley’s conception, but it is not theirs. A certain 92measure goes with protoplasm to constitute it an organism to them, and worthy of their attention. They refuse to give consideration to any mere protoplasm-shred that may not have yet ceased, perhaps, to exhibit all sign of contractility under the microscope, and demand a protoplasm-cell. ”

“ The Lymphocytes. These are small cells, as a rule approximating in size to the red blood corpuscles. Their body is occupied by a large round homogeneously stained nucleus centrally situated, whilst the protoplasm surrounds the nucleus as a concentric border. Between nucleus and protoplasm there is often found a narrow areola, which doubtless results from artificial retraction. Nucleus and protoplasm are basophil, nevertheless in many methods of staining the protoplasm possesses a much stronger affinity for the basic stain than does the nucleus. ”
